Nasce a Varsavia nel 1910 da una famiglia di tradizione socialista. Fin da piccola trascorre molto tempo con i suoi coetanei di origine ebrea, e a 5 anni è in grado di parlare yddish. Il padre è medico e fra i suoi pazienti ci sono molti ebrei poveri di cui si prende cura gratuitamente. Alla sua morte nel 1917, la comunità ebraica offre un sussidio alla famiglia in segno di gratitudine. Da ragazza Irena entra nel movimento scout e durante gli anni universitari si oppone apertamente alla discriminazione degli studenti ebrei. Entra nell’Associazione della Gioventù Polacca Democratica e nel Partito Socialista Polacco. 

Quando scoppia la Seconda Guerra Mondiale, ha 29 anni e lavora come assistente sociale per l’amministrazione comunale, dove, con il supporto del direttore del dipartimento (che per questo verrà deportato ad Auschwitz), soccorre gli ebrei oggetto di ogni tipo di discriminazione da parte dell’autorità nazista occupante. Nell’autunno del 1940, a Varsavia, viene recintato il ghetto e quasi 400.000 ebrei sono trasferiti al suo interno in condizioni igieniche precarie, aggravate dalla mancanza di cibo e medicine: si moltiplicano le epidemie e il tasso di mortalità è altissimo. In veste di infermiera, Irena riesce ad ottenere un lasciapassare: ufficialmente entra per la disinfestazione, in realtà organizza una rete di soccorso procurando cibo, generi di conforto, vestiti. Quando è nel ghetto porta la stella di David, non solo per confondersi fra la folla, ma anche in segno di solidarietà. Nel 1942 nasce l’organizzazione segreta “Consiglio per l’aiuto agli ebrei” (Zegota) e Irena ne diventa subito una tra le principali attiviste come responsabile del dipartimento infantile di Zegota con il nome in codice di Jolanta,. Alla decisione dei tedeschi di liquidare il ghetto inizia a trasferire i bambini, vestita da infermiera, nascondendoli nelle ambulanze. Spesso i piccoli vengono addormentati con i sonniferi e rinchiusi in un sacco o in una cassa per passare nella parte ariana, facendo credere agli uomini della gestapo che si tratta di morti per tifo. Dopo l’uscita dal ghetto i bambini sono raccolti in centri di assistenza, dove imparano ad adattarsi al nuovo ambiente, e poi assegnati a famiglie, orfanotrofi o conventi. Il 20 ottobre del 1943, i nazisti arrestano “Jolanta” e la torturano brutalmente per tre mesi senza riuscire a farla parlare; la condannano a morte e la trasferiscono nel terribile carcere di Pawiak. Zegota riesce, all’ultimo momento, a corrompere un generale nazista con una grossa somma di denaro per salvarla poco prima della fucilazione. Da quel momento la sua vita cambia, non può più entrare nel ghetto: deve necessariamente vivere in clandestinità, con il nome di Klara Dabrowska, perché ufficialmente è stata fucilata. Questo non le impedisce di continuare a collaborare con Zegota e aiutare gli ebrei, coordinando il salvataggio di molti bambini. Non è ancora noto esattamente quanti ne abbia salvati, ma si parla di circa 2.500. Durante l’Insurrezione di Varsavia lavora come infermiera nel Punto Sanitario e dopo la guerra entra nel Centro di Aiuto Sociale della capitale. Contribuisce a creare orfanotrofi, un Centro di Assistenza per le Madri e i Bambini in difficoltà, alcune istituzioni a sostegno delle famiglie disoccupate.


Viene perseguitata anche dai Servizi di Sicurezza comunisti: nel 1949 è arrestata e brutalmente interrogata, perchè sospettata di nascondere membri dell’Esercito Partigiano (AK); in carcere perde un bambino nato prematuramente. Dal 1948 al 1968 è iscritta al Partito Operaio Unificato Polacco, da cui esce dopo nel 1968, in segno di protesta per le repressioni contro studenti ed intellettuali, e per la campagna antisemita lanciata dal governo Nel 1980 aderisce a Solidarnosc. Negli anni del comunismo la ”Repubblica Popolare Polacca” si dimentica di lei. Nel 1999, un insegnante americano, Norman Conard, colpito dalla sua storia, fa mettere in scena dai suoi allievi del Kansas un’opera teatrale intitolata Life in a Jar (la vita in un barattolo). Il titolo si riferisce al modo con cui Irena aveva conservato i nomi dei bambini salvati, interrandoli sotto un melo del giardino, chiusi in un barattolo di vetro, con i dati dei veri genitori e di quelli adottivi, insieme ad alcuni effetti personali.


Al termine del conflitto la sua lista, consegnata ai leader della comunità ebraica, permise ai bambini di scoprire le proprie origini e ritrovare le famiglie. L’opera teatrale è stata replicata oltre duecento volte negli Stati Uniti e ha portato alla creazione della fondazione „Life in a Jar” che promuove la figura della Sendler.. La Sendler ha ricevuto numerosi premi e riconoscimenti in patria e all’estero. Nel 1965 l’istituto di Yad Vashem le ha conferito la Medaglia di ”Giusto fra le Nazioni” e nel 1991 ha ricevuto la cittadinanza onoraria di Israele. Nel 2006 l’associazione ”I figli dell’Olocausto” (Dzieci Holocaustu) insieme al Ministero degli Esteri ha dato vita al premio ”Irena Sendler” per aver reso migliore il mondo. Nel 2007 è stata candidata al Premio Nobel per la pace. Nel 2009 è stato girato il film I figli di Irena Sendler interpretato dall’attrice premio Oscar Anna Paquin. L’annuncio della sua scomparsa, il 12 maggio 2008 a Varsavia, ha suscutato grande emozione nel Paese. Di lei Marek Edelman ha detto “E’ morta una grande persona - una persona con un cuore grande, con un grande talento organizzativo, una persona che si è battuta sempre dalla parte dei deboli"
